DETERMINATION OF CARBAMAZEPINE IN PHARMACEUTICAL PREPARATIONS USING HIGH-PERFORMANCE LIQUID CHROMATOGRAPHY AND DERIVATIVE SPECTROPHOTOMETRY

Abstract A sensitive reversed, phase high-performanee liquid, ehromatographie (HPLC) and seeond derivative speetrophotometrie methods for determination of earbamazepine in tablets have been developed. In the HPLC method, earbamazepine was separated using Phenomenex C18 eolumn and aeetonitrile:water (75:25,v/v) the mobile phase system; the speed of the mobile phase flowing was 1mL/min and the deteetion was aetualized at 285 nm. Enalapril was used as an internal standard. For the seeond-derivative speetrophotometrie method, earbamazepine was determined by applying the teehnique of the "peak to peak amplitudes".The assay was linear over the eoneentration range of 0.2-2.0 Ug/mL for HPLC and 4.0-10.0 Ug/mLfor seeond derivative speetrophotometrie method. The deteetion limits ofearbamazepine were 0.055 and 1.25 Ug/mLfor HPLC and derivative speetrophotometrie methods, respeetively. The reeovery (mean±RSD) in HPLC was 99.22±0.25% and in the derivative speetrophotometry 99.05±0.25%. The proposed methods were sueeessfully applied to the determination of earbamazepine in tablets: the reeovery was of high pereentage, the aeeuraey and presieion were good.
